=== Purity and purification === TLC helps show the purity of a sample. A pure sample should only contain one spot by TLC. TLC is also useful for small-scale purification. Because the separated compounds will be on different areas of the plate, a scientist can scrape off the stationary phase particles containing the desired compound and dissolve them into an appropriate solvent. Once all the compound dissolves in the solvent, they filter out the silica particles, then evaporate the solvent to isolate the product. Big preparative TLC plates with thick silica gel coatings can separate more than 100 mg of material. TLC is useful for larger-scale purification and isolation to quickly test solvent mixtures before running flash column chromatography on a large batch of impure material. A compound elutes from a column when the amount of solvent collected is equal to 1/RF. The eluent from flash column chromatography gets collected across several containers (for example, test tubes) called fractions. TLC helps show which fractions contain impurities and which contain pure compound. Furthermore, two-dimensional TLC can help check if a compound is stable on a particular stationary phase. This test requires two runs on a square-shaped TLC plate. The plate is rotated by 90º before the second run. If the target compound appears on the diagonal of the square, it is stable on the chosen stationary phase. Otherwise, it is decomposing on the plate. === Depression === A comparative meta-analysis of 21 major antidepressants found that venlafaxine, agomelatine, amitriptyline, escitalopram, mirtazapine, paroxetine, and vortioxetine were more effective than other antidepressants, although the quality of many comparisons was assessed as low or very low. Open-label evidence also suggests potential benefit in treatment-resistant cases: in a multicenter Canadian study of adults with inadequate response to prior antidepressants (n=159), 58% achieved response and 28% remission after 8 weeks of venlafaxine (mean 260 mg/day); tolerability was generally acceptable. Venlafaxine was similar in efficacy to the atypical antidepressant bupropion; however, the remission rate was lower for venlafaxine. In a double-blind study, patients who did not respond to an SSRI were switched to either venlafaxine or another SSRI (citalopram); similar improvement was observed in both groups. Studies have not established its efficacy for use in pediatric populations. In children and adolescents with depression, venlafaxine increases the risk of suicidal thoughts or attempts. Higher doses (e.g., 225 mg and 375 mg per day) of venlafaxine are more effective than lower doses (e.g., 75 mg per day) but also cause more side effects. Studies have shown that the extended-release is superior to the immediate-release form of venlafaxine. For a long time Le Havre has exploited the strengths of its coastal location but also suffered from its relative isolation. This is why the accessibility of the city has been improved with the harbour highway A131 (E05) which links Le Havre to the A13 autoroute over Tancarville Bridge. More recently the A29 autoroute (E44) has connected Le Havre to the north of France and passes over the Normandy Bridge which makes Amiens (in the north-east) two hours away and Caen (in the south-west) one hour. The TER network was modernized with the creation of the LER line in 2001 and direct services to Fécamp in 2005. Thirteen Corail trains of the Paris–Le Havre line link Le Havre station with Bréauté-Beuzeville, Yvetot, Rouen and Paris Saint-Lazare station. In addition there is a TGV daily service to Le Havre: it has connected the city to Marseille since December 2004 serving Rouen, Mantes-la-Jolie, Versailles, Massy, Lyon, Avignon, Aix-en-Provence, and Saint Charles station in Marseille. === Physiology === Potassium is the most abundant intracellular cation. About 98% of the body's potassium is found inside cells, with the remainder in the extracellular fluid, including the blood. In healthy individuals, homeostasis is maintained when cellular uptake and kidney excretion naturally counterbalance a patient's dietary intake of potassium. ==== Cell-based scheme of coagulation ==== A newer model of coagulation mechanism explains the intricate combination of cellular and biochemical events that occur during the coagulation process in vivo. Along with the procoagulant and anticoagulant plasma proteins, normal physiologic coagulation requires the presence of two cell types for formation of coagulation complexes: cells that express tissue factor (usually extravascular) and platelets. The coagulation process occurs in two phases. First is the initiation phase, which occurs in tissue-factor-expressing cells. This is followed by the propagation phase, which occurs on activated platelets. The initiation phase, mediated by the tissue factor exposure, proceeds via the classic extrinsic pathway and contributes to about 5% of thrombin production. The amplified production of thrombin occurs via the classic intrinsic pathway in the propagation phase; about 95% of thrombin generated will be during this second phase.
